Takara’s new DNA ligation Kit LONG is a powerful
tool for cloning DNA fragments from 2 kb to over
10 kb in length. The optimized
ligase/buffer system enables ligation
of long fragments without difficult
techniques and special expertise. Ligation Kit
LONG gave cloning efficiencies about 50X higher
than other commercially available DNA ligation
kits when inserting high molecular weight DNA
(up to 18 kb) into a vector (see figure).

DNA Ligation Kits. Hind III-digested DNA fragments of varying size (2 kb,
4 kb, 10 kb and 18 kb) were ligated into the cloning vector pUC118/Hind
III/BAP using the Ligation Kit LONG (Lig LONG) and several other “I used Takara’s DNA Ligation Kit LONG to
